I have a few different forms of spyware from one source how do i get rid of them?

I have a yellow triangle that pops up every twenty seconds on my taskbar, i have a red window that pops up to show an infected file and says the threat is CoolWebSearch, and i also have a windows privacy popup that says i have TrojanDownloader.XS. The yellow triangle takes me to to a website that has the top rated spyware removal tools please help me

Spybot,I run it every week.
