How do I get rid of adware and spyware that my antivirus scan can’t find?

I keep getting a pop up every time I turn on my laptop. I also keep hearing a a guy or girl saying “Congratulations. You’ve just won whatever. Just click here…” No window pops up when the person starts talking. Just a few minutes ago I started getting the Critical Error beep and a small icon on the lower right keeps saying “Warning, You have a security problem!”. I think it might be a WinWeb spyware. I also had AntiVirus 2010 and manually removed it. I don’t know if I removed everything that I needed to remove.
I have McAfee antivirus. It doesn’t find anything when I run the scan. Any suggestions??

1. Download threatfire. It might detect a virus McAfee can’t detect.

2. Download google toolbar. It has a popup blocker.

3. Download spybot search & destroy. You might have some sort of spyware on your computer.
